545 Importance of CD40 expression on s. aureus superantigen responses in human keratinocytes

چکیده

S. aureus produces secreted virulence factors, including superantigens (SAgs), that activate the host immune response and cause tissue damage. Little is known about what receptors SAgs utilize on epithelial cells, keratinocytes (KC) in particular. CD40 has recently been proposed as a potential SAg receptor for TSST-1 vaginal cells. To address relevance of KC, we utilized CRISPR/Cas9 to knockout (KO) N/TERT2G KC cell line established mock-transfected wild type (WT) clonal KO. KO was confirmed by flow cytometry, sequence analysis. When differentiated, had significantly diminished barrier (38-48% reduction compared with WT, p=0.009-0.03), measured transepithelial electrical resistance (TEER). We assessed how impacted responsiveness purified SAgs. Clones were treated ten individually qPCR inflammatory cytokines IL-6, IL-8, TNFa conducted. In WT SElQ, SElU, induced most impressive increases, fold increase IL-8 transcripts (over sham-treated controls) 95.5 23.0 SElU 15.0 TSST-1. contrast, clone an attenuated SElQ (34.0; 2.8-fold less), (3.8; 6.0-fold TSTT-1 (2.0; 7.5-fold less). This suggests expression important optimal responses, but it unlikely necessary act KC. Additionally, may have specificity only certain previously shown some make much more permissive viral infections. Ongoing studies are investigating role vaccinia virus herpes simplex virus-1.

Role of Staphylococcal Superantigen in Atopic Dermatitis: Influence on Keratinocytes

Staphylococcus aureus may perform an crucial function in atopic dermatitis (AD), via the secretion of superantigens, including staphylococcal enterotoxins (SE) A or B, and toxic shock syndrome toxin-1 (TSST-1). Dysregulated cytokine production by keratinocytes (KCs) upon exposure to staphylococcal superantigens (SsAgs) may be principally involved in the pathophysiology of AD. We hypothesized th...

Study effects of Staphylococcus aureus superantigen C on production of C- reactive protein in rat

Background and purpose: Recently, microbial super antigens have been reported in blood and synovial fluid of rheumatoid arthritis patients. Whether or not, the presence of these super antigens could provoke the induction of serologic diagnostic factors, including CrP in patients? the purpose of this study was to production and purification one of the super antigen C of Staphylococcus aureus and...

Signal transduction and nuclear responses in Staphylococcus aureus-induced expression of human beta-defensin 3 in skin keratinocytes.

The human beta-defensin 3 (hBD-3) is an inducible epithelial peptide antibiotic that has potent antistaphylococcal activity. Infection of skin epithelial cells with viable Staphylococcus aureus, a common skin pathogen, induces increased gene expression of hBD-3 and other antimicrobial peptides. The aim of this study was to identify signaling pathways and nuclear responses that contribute to the...

Undetectable CD40 ligand expression on T cells and low B cell responses to CD40 binding agonists in human newborns.

IgG, IgA, and IgE production by newborn B cells is limited both in vivo and in vitro in various activation conditions, whereas IgM production is readily detectable. It has been suggested that the Ig heavy chain switch inability could be the consequence of T and B cell immaturity. As the interaction between CD40 (expressed on B cells) and its ligand CD40-L (expressed on activated T cells) trigge...
